To speak in behalf of someone is to plead that person's case. To speak on behalf of someone is to speak in the person's stead. These two sentences illustrate the correct usage: "I was about to be fired, but Myrna spoke to the boss in my behalf," and "Addressing the shareholders, the chairman spoke on behalf of all the company's employees."
