Determination of Rf (retention factor) value of amino acid by thin-layer chromatography (TLC) and identification of the amino acid present.

THEORY

TLC is a special type adsorption chromatography in which a thin layer of the adsorbent on a flat surface is utilized instead of column of adsorbent. Elution and development of the chromatogram is accomplished by capillary movement of the solvent up the thin layer of the adsorbent.

In this technique, the material to be identified or separated is dissolved in a suitable solvent. It is spotted near one end of the plate covered with a thin layer of silica gel or alumina containing some binding material such as calcium sulphate.
