Chapter 27. Culturing Bacteria

Equipment and Materials

You’ll need the following items to complete this lab session. (The standard kit for this book, available from www.thehomescientist.com, includes the items listed in the first group.)

Materials from Kit

  • Goggles

  • Coverslip

  • Inoculating loop

  • Microscope slide, flat

  • Pipettes

  • Stain, Gram’s iodine

  • Stain, Hucker’s crystal violet

  • Stain, safranin O

  • Stirring rod

  • Test tube rack

Materials You Provide

  • Gloves

  • Alcohol (ethanol or isopropanol)

  • Alcohol lamp, candle, or butane lighter

  • Agar Petri dishes (from preceding lab)

  • Agar slant tubes (from preceding lab)

  • Bleach, chlorine laundry

  • Broth culturing tubes (from preceding lab)

  • Clock or watch with second hand

  • Large container with lid (for bleach bath)

  • Microscope

  • Mixed bacteria culture ...
