This chapter describes the mechanism-based screening for the discovery of novel antifungals. Antifungal agents were identified by the ability to inhibit fungal growth in agar solidified media. There are a number of variations on this basic assay design, including the Walksman streak assay, agar overlay method, and disk or agar plug diffusion assay. All these methods resemble one another in that the identification of the antibiotic substance depends upon its ability to inhibit the growth of the detection organism while diffusing through an agar solidified medium.
